Functional Brain Networks and Cognitive Disorders in Older People

Abstract
Alzheimer¡¯s disease (AD) is the most common cause of neurodegenerative disease. The AD-related neuropathology can disrupt neural activities at various levels such as molecular pathways, synapses, neuronal sub-populations and local circuits in specific brain regions, as well as higher-order neural networks. A mountain of evidence supported the idea of AD as disconnection syndrome. Therefore, network-based approach is critical to understand AD pathology-related brain change and neural substrates of clinical manifestations. In this review, background history of application of network-based approach to neuroscience field and basic concept of graph theory were briefly introduced. Recent studies of functional brain networks in cognitive disorders including AD and mild cognitive impairment were summarized. Methodological issues and limitations of previous studies, as well as future perspectives, were discussed.
